WNT5B是WNT基因家族的重要成员之一,该家族编码一组分泌型糖蛋白,在胚胎发育、细胞增殖、分化及组织稳态中发挥关键作用。WNT5B属于非经典WNT信号通路(WNT/β-catenin非依赖通路),主要通过激活平面细胞极性(PCP)通路和WNT/Ca²⁺通路调控细胞迁移、极性建立和细胞命运决定。其表达产物WNT5B蛋白通过结合卷曲受体(Frizzled)和共受体(如ROR2/RYK)传递信号,在骨骼发育、脂肪代谢和血管形成等过程中具有特异性功能。WNT5B突变可能导致信号传导异常,与多种疾病相关,例如功能丧失性突变可能引起骨骼发育障碍(如Robinow综合征),而过度激活则与癌症(如结直肠癌、乳腺癌)和代谢性疾病(如肥胖、2型糖尿病)的发生发展密切相关。当WNT5B过表达时,会异常激活下游JNK和NF-κB等促炎通路,促进肿瘤细胞侵袭转移;同时抑制脂肪细胞分化,导致胰岛素抵抗。反之,低表达可能破坏组织再生能力,如影响骨修复或血管网络形成。WNT基因家族的共性在于均依赖脂质修饰(棕榈酰化)实现分泌,并通过保守的半胱氨酸残基与受体相互作用。目前"非经典WNT通路"(non-canonical Wnt pathway)的中文译法存在争议,部分文献直译为"非规范通路"。该家族成员功能高度分化,如WNT5A与WNT5B虽同属非经典亚型,但时空表达模式和具体生物学效应存在差异。

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 94% and 80% amino acid identity to the mouse Wnt5b protein and the human WNT5A protein, respectively. Alternative splicing of this gene generates 2 transcript variants. [provided by RefSeq, Jul 2008]
