VDR(维生素D受体,Vitamin D Receptor)是核受体超家族(NR1I1亚家族)的成员,主要介导维生素D的生物学功能。其表达产物是一种配体依赖的转录因子,在肠道、肾脏、骨骼、免疫细胞等多种组织中广泛分布,通过与活性维生素D(1,25-二羟维生素D3)结合形成复合物,调控靶基因的转录。VDR的核心功能包括维持钙磷代谢平衡(促进肠道钙吸收和肾脏钙重吸收)、调节骨骼发育(促进成骨细胞分化)、参与免疫调节(抑制炎症因子如TNF-α、IL-6的释放)以及调控细胞增殖与分化(如抑制某些癌细胞生长)。该基因的主要作用位点包括靶基因启动子区的维生素D反应元件(VDRE),通过招募共激活因子或共抑制因子来调控下游基因(如钙结合蛋白Calbindin、骨基质蛋白Osteocalcin等)的表达。VDR突变可导致遗传性维生素D抵抗性佝偻病(HVDRR),表现为低钙血症、骨骼畸形和脱发,其功能丧失性突变会完全阻断维生素D信号通路。VDR与多种疾病相关:自身免疫病(如多发性硬化症、1型糖尿病)中其表达常降低;而在前列腺癌、乳腺癌中VDR信号通路异常可能促进肿瘤进展。VDR过表达可增强抗炎效应和抑制肿瘤生长,但可能导致高钙血症;低表达则与骨质疏松、免疫力下降相关。VDR基因家族(核受体NR1亚家族)的共性包括:具有高度保守的DNA结合域(DBD)和配体结合域(LBD),以二聚体形式(常与RXR受体配对)结合DNA,并受脂溶性激素/维生素调控。该家族成员普遍参与代谢、发育和稳态维持。
