THRA基因编码甲状腺激素受体α(thyroid hormone receptor alpha),属于核受体超家族(nuclear receptor superfamily)中的甲状腺激素受体家族(THR family)。该家族包括THRA和THRB两个亚型,均通过与甲状腺激素(T3/T4)结合调控靶基因转录。THRA主要在心脏、骨骼肌、大脑和骨骼中表达,其蛋白产物存在两种亚型:TRα1(功能性受体)和TRα2(不能结合激素的变体)。正常功能下,TRα1与激素结合后形成二聚体,结合DNA上的甲状腺激素反应元件(TREs)调控细胞增殖、分化及代谢相关基因(如MYH7、DIO2等)。该基因突变可导致甲状腺激素抵抗综合征(RTHα),表现为便秘、生长迟缓、骨骼发育异常等,典型突变如p.A263V会损害激素结合能力。过表达TRα1会抑制促甲状腺激素(TSH)分泌,导致类似甲亢的心动过速;而表达不足则引发心肌收缩力下降。该基因与阿尔茨海默病相关,其表达降低可能影响神经元存活。THRA与THRB功能存在部分冗余,但THRA更主导调控心脏和中枢神经系统发育。基因家族共性包括:均含有DNA结合域(DBD)和配体结合域(LBD),通过激素依赖性或非依赖性方式调控转录,参与发育和能量代谢调控。

The protein encoded by this gene is a nuclear hormone receptor for triiodothyronine. It is one of the several receptors for thyroid hormone, and has been shown to mediate the biological activities of thyroid hormone. Knockout studies in mice suggest that the different receptors, while having certain extent of redundancy, may mediate different functions of thyroid hormone. Alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]
