TGM2(组织型转谷氨酰胺酶2,tissue transglutaminase 2)是一种多功能酶,属于转谷氨酰胺酶家族(transglutaminase family),该家族共有8个成员(TGM1-TGM7及FXIII-A),其共性是能够催化蛋白质间交联反应,通过形成ε-(γ-谷氨酰)赖氨酸键(一种共价键)来稳定蛋白质结构或参与细胞信号传导。TGM2在钙离子依赖下发挥主要功能,但也存在不依赖酶活性的作用方式。其表达产物广泛分布于细胞质、细胞核、线粒体及细胞外基质中,主要作用位点包括参与细胞凋亡(程序性细胞死亡)、细胞黏附、伤口愈合、炎症反应及能量代谢等过程。该基因突变可能导致酶活性异常,与多种疾病相关,例如乳糜泻(celiac disease)患者体内TGM2会错误修饰麸质蛋白,引发自身免疫反应;某些神经退行性疾病(如亨廷顿病)中也观察到TGM2介导的异常蛋白聚集。过表达TGM2会促进纤维化疾病(如肝纤维化、肾纤维化)的进展,因其加剧细胞外基质沉积;还可能与肿瘤转移相关,通过增强癌细胞侵袭能力。相反,降低TGM2表达可缓解这些病理过程,但可能影响组织修复功能,因其正常水平对伤口愈合至关重要。在癌症中,TGM2表现出双重角色:在某些肿瘤中过表达促进生存(如乳腺癌),而在其他情况下却抑制肿瘤(如黑色素瘤),这种差异可能与细胞微环境有关。该基因还参与自噬(细胞自我降解回收过程)调控,并与NF-κB等信号通路相互作用,影响炎症因子释放。

Transglutaminases are enzymes that catalyze the crosslinking of proteins by epsilon-gamma glutamyl lysine isopeptide bonds. While the primary structure of transglutaminases is not conserved, they all have the same amino acid sequence at their active sites and their activity is calcium-dependent. The protein encoded by this gene acts as a monomer, is induced by retinoic acid, and appears to be involved in apoptosis. Finally, the encoded protein is the autoantigen implicated in celiac disease. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]
