TFAP4(转录因子AP-4)属于碱性螺旋-环-螺旋(bHLH)转录因子家族,这类蛋白通过螺旋结构域结合DNA并调控基因表达。TFAP4通过识别特定DNA序列(E-box元件,如CAGCTG)调控下游靶基因,参与细胞周期、增殖、分化及凋亡等过程。其表达产物在胚胎发育、神经形成和免疫调节中起关键作用,尤其在神经嵴细胞迁移和肠上皮稳态维持中表现突出。TFAP4突变可能导致功能丧失或异常激活,与多种癌症相关(如结直肠癌、胃癌、神经母细胞瘤),突变后可能破坏细胞周期调控,促进肿瘤侵袭。过表达TFAP4会加速细胞增殖并抑制凋亡,可能通过上调Cyclin D1等促癌基因驱动肿瘤发生;而表达降低则可能引发分化缺陷或细胞周期停滞。该基因属于AP-4转录因子家族(含TFAP4/TFAP2等),家族共性为通过保守的bHLH结构域二聚化并结合DNA,调控发育相关基因。在疾病中,TFAP4过表达常见于恶性肿瘤,与不良预后相关;而敲除实验显示其缺失可能导致神经系统异常或肠道屏障功能障碍。目前"转录因子AP-4"的中文译名存在争议,部分文献保留英文原名(TFAP4)以避免混淆。
