TCL1A(T细胞白血病/淋巴瘤1A)基因属于TCL1基因家族,该家族还包括TCL1B和MTCP1等成员,主要参与调控淋巴细胞(尤其是T细胞和B细胞)的增殖与存活。TCL1基因家族的共性是通过激活AKT信号通路(一种促进细胞生长和存活的分子途径)来发挥作用。TCL1A蛋白通常与AKT结合,增强其磷酸化(一种激活方式),从而抑制细胞凋亡(程序性死亡)并促进细胞分裂。该基因主要在胸腺、脾脏和淋巴结等免疫组织中表达,是淋巴细胞发育的关键调节因子。TCL1A的异常激活或突变与多种血液系统恶性肿瘤密切相关,例如T细胞幼淋巴细胞白血病(T-PLL)和B细胞慢性淋巴细胞白血病(B-CLL)。在这些疾病中,TCL1A常因染色体易位(如t(14;14)或t(X;14))或基因扩增导致过表达,进而过度激活AKT通路,驱动癌细胞不受控增殖。若TCL1A表达降低,则可能削弱淋巴细胞存活能力,导致免疫功能异常。此外,TCL1A过表达还会影响其他基因,如上调抗凋亡蛋白BCL2的表达,同时抑制肿瘤抑制因子(如PTEN)的功能。研究还发现,TCL1A转基因小鼠(人为增加该基因的小鼠模型)会自发发展白血病,印证其致癌潜力。目前针对TCL1A相关疾病的治疗策略包括靶向AKT通路抑制剂,如MK2206。需要注意的是,TCL1A的异常表达可能干扰正常免疫监视功能,增加感染或自身免疫疾病风险。该基因的调控机制涉及表观遗传修饰(如DNA甲基化)和微小RNA(如miR-34a)的负调控。

Overexpression of the TCL1 gene in humans has been implicated in the development of mature T cell leukemia, in which chromosomal rearrangements bring the TCL1 gene in close proximity to the T-cell antigen receptor (TCR)-alpha (MIM 186880) or TCR-beta (MIM 186930) regulatory elements (summarized by Virgilio et al., 1998 [PubMed 9520462]). In normal T cells TCL1 is expressed in CD4-/CD8- cells, but not in cells at later stages of differentiation. TCL1 functions as a coactivator of the cell survival kinase AKT (MIM 164730) (Laine et al., 2000 [PubMed 10983986]).[supplied by OMIM, Jul 2010]
