TAL1(T-cell acute lymphocytic leukemia protein 1,也称SCL或干细胞白血病基因)是一种在造血系统中起关键作用的转录因子基因,属于碱性螺旋-环-螺旋(bHLH,basic helix-loop-helix)蛋白家族。该家族成员通常通过形成二聚体结合DNA,调控靶基因表达,参与细胞分化、增殖等过程。TAL1在胚胎造血、血管形成及T细胞发育中发挥核心作用,其表达产物通过与其他bHLH蛋白(如E2A、HEB)结合形成复合物,激活或抑制下游基因(如GATA1、MYB)的表达,从而影响红系、髓系及淋系细胞的分化。TAL1的典型作用位点为DNA上的E-box序列(CANNTG),通过调控造血相关基因网络维持正常造血功能。TAL1基因突变(如染色体易位t(1;14)或t(1;7))会导致其异常表达,破坏造血细胞分化平衡,约60%的T细胞急性淋巴细胞白血病(T-ALL)病例中存在TAL1过表达,这与白血病发生密切相关。若TAL1过表达,会过度激活增殖信号通路(如NOTCH1),抑制细胞凋亡,促进恶性转化;而敲低TAL1则导致造血干细胞自我更新能力下降和红系分化障碍。TAL1基因家族(bHLH家族)的共性包括:依赖螺旋-环-螺旋结构域形成同源或异源二聚体,广泛参与细胞命运决定和器官发育。目前“TAL1”的中文译名存在争议,部分文献保留英文缩写或使用“T细胞急性淋巴细胞白血病蛋白1”。
