SULF2(硫酸酯酶2)是一种细胞外硫酸酯酶,属于硫酸酯酶基因家族(Sulfatase家族),该家族成员共同特点是能够水解硫酸酯键,参与调控硫酸乙酰肝素蛋白聚糖(HSPGs)的硫酸化修饰。SULF2通过选择性去除HSPGs上的6-O-硫酸基团,影响多种生长因子(如FGF、VEGF、Wnt等)的信号传导,从而参与细胞增殖、迁移、分化和血管生成等生物学过程。SULF2在胚胎发育、组织修复和肿瘤发生中起重要作用,其表达异常与多种癌症(如肝癌、乳腺癌、胶质瘤)密切相关。SULF2过表达会增强生长因子信号通路活性,促进肿瘤细胞侵袭和转移,同时刺激血管生成,而降低其表达则可能抑制肿瘤进展。SULF2突变可能导致其酶活性丧失,影响HSPGs的硫酸化模式,进而扰乱细胞微环境和信号传导。此外,SULF2在神经系统发育和肝纤维化中也发挥作用,其表达水平变化可能影响神经细胞迁移或胶原沉积。该基因与SULF1高度同源,两者共同调节HSPGs的硫酸化状态,但SULF2更倾向于在病理条件下(如癌症)被激活。研究SULF2有助于开发针对癌症和纤维化疾病的靶向治疗策略。

Heparan sulfate proteoglycans (HSPGs) act as coreceptors for numerous heparin-binding growth factors and cytokines and are involved in cell signaling. Heparan sulfate 6-O-endosulfatases, such as SULF2, selectively remove 6-O-sulfate groups from heparan sulfate. This activity modulates the effects of heparan sulfate by altering binding sites for signaling molecules (Dai et al., 2005 [PubMed 16192265]).[supplied by OMIM, Mar 2008]
