SPP1(Secreted Phosphoprotein 1,分泌磷蛋白1)也称为骨桥蛋白(Osteopontin),是一种多功能糖基化磷蛋白,属于SIBLING(Small Integrin-Binding Ligand N-linked Glycoprotein,小整合素结合N-连接糖蛋白)基因家族成员。该家族共性是通过整合素(细胞表面受体)和CD44(细胞黏附分子)介导细胞间或细胞与基质的相互作用,参与组织矿化、炎症和肿瘤发生等过程。SPP1的生物学功能广泛,其表达产物通过结合整合素(如αvβ3)和CD44受体,调控细胞黏附、迁移、增殖及免疫应答。它在骨组织中促进羟基磷灰石结晶沉积,参与骨重塑;在免疫系统中激活巨噬细胞和T细胞,增强炎症反应;在肿瘤微环境中则促进癌细胞转移和血管生成。SPP1的主要作用位点包括骨骼、肾脏、免疫细胞及多种上皮组织。突变可能导致功能异常,如SPP1基因敲除小鼠表现为骨密度降低和伤口愈合延迟,而人类某些SPP1变异与肾结石、动脉粥样硬化和自身免疫疾病(如多发性硬化症)相关。过表达SPP1会加剧炎症性疾病(如类风湿关节炎)和肿瘤进展(如乳腺癌、肝癌),因其促进细胞侵袭和免疫逃逸;而降低表达则可能抑制组织纤维化(如肺纤维化)但增加骨质疏松风险。在癌症中,SPP1过表达通过激活PI3K-AKT(促生存信号通路)和NF-κB(炎症通路)促进转移,并与不良预后相关。其表达水平受miRNA(如miR-181a)和表观遗传修饰(如DNA甲基化)调控。目前SPP1作为生物标志物或治疗靶点在多种疾病中被研究,例如抗SPP1抗体在肿瘤免疫治疗中的潜在应用。

The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene.
