SPARC(Secreted Protein Acidic and Rich in Cysteine,富含半胱氨酸的酸性分泌蛋白)是一种多功能糖蛋白,属于SPARC基因家族(也称为骨连接素家族或BM-40家族),该家族成员具有调节细胞外基质(ECM,细胞周围的蛋白质网络)组装和细胞-基质相互作用的功能。SPARC主要由成骨细胞、成纤维细胞和内皮细胞分泌,在组织发育、伤口修复和骨形成中起关键作用。其生物学功能包括抑制细胞增殖、调节ECM沉积(如胶原蛋白和纤连蛋白的组装)、影响细胞黏附与迁移,并通过结合钙离子参与矿化过程(如骨骼和牙齿的形成)。主要作用位点为细胞外基质及细胞表面受体(如整合素和生长因子受体)。SPARC基因突变可能导致ECM结构异常,与骨质疏松、纤维化疾病和某些癌症(如乳腺癌、胶质母细胞瘤)的进展相关。突变可能削弱其抑制肿瘤生长的能力或加剧ECM紊乱。在疾病中,SPARC的表达具有双重性:在部分肿瘤中高表达促进转移(通过增强细胞迁移),而在其他情况下(如结肠癌)低表达与不良预后相关。过表达SPARC可能抑制血管生成(通过拮抗VEGF信号通路)从而限制肿瘤生长,但也会加速纤维化疾病(如肺纤维化)的ECM过度沉积;低表达则可能导致组织修复缺陷或骨质疏松。SPARC家族成员的共性是含有保守的ECM结合域(如FS和EC结构域)和钙离子结合位点,均参与细胞分化与组织重塑。专业术语解释:细胞外基质(ECM)是支撑细胞的蛋白质网络;整合素是细胞表面受体,介导细胞与ECM的黏附;血管生成指新血管形成。目前中文对"SPARC"的翻译尚有争议,部分文献直译为"SPARC蛋白"或沿用旧称"骨连接素(osteonectin)"。

This gene encodes a cysteine-rich acidic matrix-associated protein. The encoded protein is required for the collagen in bone to become calcified but is also involved in extracellular matrix synthesis and promotion of changes to cell shape. The gene product has been associated with tumor suppression but has also been correlated with metastasis based on changes to cell shape which can promote tumor cell invasion.
