SOS2(Son of Sevenless homolog 2)是SOS基因家族的重要成员,该家族还包括SOS1,它们编码鸟苷酸交换因子(GEFs),主要功能是激活RAS蛋白(一类调控细胞生长的关键分子开关)。SOS2通过促进RAS从非活性的GDP结合状态转变为活性的GTP结合状态,从而激活下游的MAPK信号通路(控制细胞增殖、分化的关键通路)。其表达产物主要作用于细胞膜附近,与生长因子受体(如EGFR)结合后传递信号。突变可能导致SOS2功能异常,例如过度激活RAS-MAPK通路,与努南综合征(Noonan syndrome,一种发育障碍疾病)和某些癌症(如肺癌、结肠癌)相关。若SOS2过表达,会持续激活RAS信号,导致细胞不受控增殖;而表达降低则可能影响组织发育或修复。SOS基因家族的共性是通过保守的CDC25结构域(负责催化RAS激活的区域)和PH结构域(帮助定位到细胞膜)发挥作用。目前“Son of Sevenless”的中文译名“七号之子”存在争议,部分文献直接使用英文原名。
