SNAP25(突触体相关蛋白25,英文全称Synaptosomal-associated protein 25)是一种重要的神经元特异性蛋白,属于SNARE蛋白家族(可溶性N-乙基马来酰亚胺敏感因子附着蛋白受体家族)。SNARE家族的共性是其成员通过形成复合体介导囊泡与靶膜的融合,从而调控神经递质释放和细胞内膜运输。SNAP25主要表达于突触前膜,与Syntaxin和VAMP(囊泡相关膜蛋白)共同形成SNARE复合体,驱动突触小泡与细胞膜融合,完成神经递质(如乙酰胆碱、多巴胺等)的释放,对神经信号传递至关重要。该基因的突变或表达异常会直接影响突触功能,例如某些错义突变(如I67N、V48F)可能导致复合体稳定性下降,引发神经系统疾病。SNAP25与多种疾病相关,包括注意力缺陷多动症(ADHD)、精神分裂症和阿尔茨海默病——在ADHD患者中常见该基因的过表达,导致多巴胺能信号过度活跃;而在阿尔茨海默病中,SNAP25可能被β-淀粉样蛋白破坏,引发突触功能障碍。若该基因过表达,可能增强神经递质释放效率,但过度激活会引发癫痫或运动失调;反之,表达降低会导致认知障碍或神经肌肉传导异常(如肉毒杆菌毒素通过切割SNAP25抑制其功能,引起肌肉麻痹)。此外,SNAP25还参与胰岛素分泌和免疫调节等非神经元功能,其旁系同源基因(如SNAP23)在非神经组织中发挥类似膜融合作用。该基因的调控异常可能通过影响其他SNARE成员(如Syntaxin1A)的活性产生级联效应,进一步干扰细胞分泌途径。
Synaptic vesicle membrane docking and fusion is mediated by SNAREs (soluble N-ethylmaleimide-sensitive factor attachment protein receptors) located on the vesicle membrane (v-SNAREs) and the target membrane (t-SNAREs). The assembled v-SNARE/t-SNARE complex consists of a bundle of four helices, one of which is supplied by v-SNARE and the other three by t-SNARE. For t-SNAREs on the plasma membrane, the protein syntaxin supplies one helix and the protein encoded by this gene contributes the other two. Therefore, this gene product is a presynaptic plasma membrane protein involved in the regulation of neurotransmitter release.
