SMURF2(SMAD ubiquitination regulatory factor 2)属于HECT型E3泛素连接酶家族,该家族成员通过泛素-蛋白酶体系统调控蛋白质降解。SMURF2的主要功能是介导靶蛋白的泛素化修饰(即给蛋白质贴上“降解标签”),其核心作用位点是TGF-β/SMAD信号通路中的SMAD1/2/3和SMAD7,通过泛素化这些信号分子调控细胞分化、增殖和凋亡。该基因还作用于非SMAD靶点如RhoA(影响细胞骨架)、PAR4(促凋亡蛋白)和TβRI(TGF-β受体)。突变会导致功能异常,如RING结构域突变会丧失泛素连接酶活性,与肿瘤转移、纤维化和心血管疾病相关。SMURF2过表达时过度降解SMAD蛋白会抑制TGF-β信号传导,促进肿瘤发生;同时通过稳定RhoA加剧癌细胞迁移。表达不足则导致SMAD蛋白积累,引发肺纤维化、动脉硬化等疾病。其双重调控特性体现在:对SMAD7的泛素化抑制TGF-β信号,但对TβRI的降解又激活该通路。该基因属于SMURF基因家族(含SMURF1/2),家族共性为含有C2膜定位域、WW蛋白相互作用域和HECT催化域,通过识别特定底物调控细胞信号网络。在癌症中,SMURF2既可作抑癌基因(如降解致癌蛋白Ras),又可作促癌因子(如促进EMT上皮间质转化),这种矛盾功能与组织微环境相关。最新研究发现其通过调控DNA损伤修复蛋白BRCA1影响化疗敏感性,且与自噬过程有交叉作用。
