SFN(Stratifin,也称为14-3-3σ)属于14-3-3蛋白家族,这是一个高度保守的调控蛋白家族,其成员通过结合磷酸化靶蛋白参与多种细胞过程,如信号转导、细胞周期调控、凋亡和应激反应。14-3-3蛋白家族的共性是通过形成同源或异源二聚体,识别特定的磷酸化丝氨酸/苏氨酸基序,从而调节靶蛋白的活性、稳定性或亚细胞定位。SFN主要在表皮细胞中高表达,其生物学功能包括维持上皮细胞稳态、参与DNA损伤修复和细胞周期阻滞(如G2/M期检查点调控)。它通过结合并稳定p53、CDC25C等关键蛋白,抑制细胞异常增殖。SFN的突变或表观遗传沉默(如启动子甲基化)常见于多种癌症(如乳腺癌、胃癌),导致其功能丧失,削弱细胞周期检查点,促进基因组不稳定性及肿瘤进展。过表达SFN可诱导细胞周期停滞或凋亡,抑制肿瘤生长,但可能影响正常上皮更新;而低表达则削弱DNA损伤应答,增加癌变风险。此外,SFN还参与炎症反应和上皮-间质转化(EMT),其表达水平变化可能影响TGF-β等信号通路。在临床中,SFN的甲基化状态可作为癌症早期诊断标志物,而其调控机制为靶向治疗提供潜在方向。
