RPH3A(Rabphilin-3A)属于Rab效应蛋白家族,主要参与神经递质释放的调控。该基因编码的蛋白质通过与Rab3A(一种小GTP酶)结合,调节突触小泡的锚定和融合过程,从而影响神经递质(如多巴胺、谷氨酸等)的释放效率。RPH3A在神经元突触前膜高度表达,其功能依赖于钙离子信号,并与突触结合蛋白(syntaxin)、SNAP-25等SNARE复合体组分相互作用。该基因家族(Rab效应蛋白家族)的共性是通过结合Rab GTP酶参与膜运输和囊泡 trafficking(运输)。若RPH3A发生功能丧失性突变,可能导致突触传递异常,与神经系统疾病如精神分裂症、自闭症谱系障碍相关。研究发现其表达下调会降低突触可塑性,而过表达可能增强神经递质释放但破坏释放精确性。某些精神疾病患者脑组织中观察到RPH3A表达异常,提示其参与突触功能紊乱的病理过程。该基因还通过影响多巴胺能神经元活动,间接调控运动协调性。需要注意的是,"Rabphilin-3A"在中文文献中偶被直译为"兔菲林-3A",但"RPH3A"或"Rabphilin-3A"更为通用。
