RPA1(Replication Protein A1)是复制蛋白A(RPA)复合体的核心亚基,属于RPA基因家族。RPA是由RPA1、RPA2和RPA3组成的异源三聚体,在DNA代谢过程中发挥关键作用,包括DNA复制、修复和重组。RPA1是其中最大的亚基,直接结合单链DNA(ssDNA),通过其寡核苷酸/寡糖结合结构域(OB-fold)稳定ssDNA并招募其他修复因子。RPA复合体作为"守护者"防止ssDNA降解或形成二级结构,并为DNA聚合酶提供平台。RPA1还参与细胞周期检查点调控,通过ATR激酶通路传递DNA损伤信号。该基因突变会导致基因组不稳定性,与多种癌症(如乳腺癌、卵巢癌)和早衰综合征相关。RPA1过表达常见于恶性肿瘤,可能促进肿瘤细胞增殖和耐药性;而表达降低则导致DNA损伤修复缺陷,增加细胞对辐射或化疗的敏感性。RPA基因家族成员均含有OB-fold结构域,具有保守的ssDNA结合能力,但各亚基在结合亲和力和功能调控上存在差异。RPA1的N端结构域还参与蛋白质相互作用,这是区别于其他家族成员的特征。研究表明,RPA1磷酸化状态(如Ser33位点)可调节其与DNA及其他修复因子的结合动态,影响同源重组修复效率。在端粒维持中,RPA1与端粒酶协同作用,其异常表达可能导致端粒功能障碍。某些病毒(如HPV)会劫持RPA1以完成自身基因组复制,这成为抗病毒治疗的潜在靶点。针对RPA1的小分子抑制剂正在开发中,旨在增强肿瘤细胞对DNA损伤剂的敏感性。
