RELA基因编码转录因子p65,是NF-κB(核因子κB)蛋白家族的核心成员之一。NF-κB家族包含五个亚基(RELA/p65、RELB、c-REL、NFKB1/p50、NFKB2/p52),它们通过形成同源或异源二聚体调控基因表达,在免疫应答、炎症反应、细胞增殖和凋亡等生理过程中发挥关键作用。RELA/p65具有转录激活域,能直接结合DNA并招募其他转录辅因子,而其他家族成员如p50缺乏转录激活域,常需与RELA形成异源二聚体(如经典的p65-p50二聚体)才能激活靶基因。RELA的活性受IκB蛋白的抑制,当细胞受到炎症因子(如TNF-α)、病原体或应激刺激时,IκB被降解,释放RELA进入细胞核调控靶基因(如IL-6、TNF-α、COX-2等)。RELA的功能突变(如DNA结合域突变)可能导致NF-κB信号通路异常激活或抑制,与多种疾病相关:过度激活会促进慢性炎症(如类风湿性关节炎)、癌症(如淋巴瘤、乳腺癌)和自身免疫病,而功能缺失可能增加感染风险或导致免疫缺陷。RELA过表达会增强促炎因子和抗凋亡基因的转录,加速肿瘤生长和转移,同时可能抑制其他转录因子(如AP-1)的活性;低表达则可能削弱免疫防御,导致组织修复障碍。此外,RELA在发育中调控神经形成和肝细胞稳态,其条件性敲除小鼠会出现肝细胞凋亡和胚胎致死。该基因的调控异常还与阿尔茨海默病、糖尿病并发症等慢性病相关。药物开发中,靶向RELA的抑制剂(如硼替佐米)被用于治疗多发性骨髓瘤等NF-κB依赖型癌症。
