RBP4(视黄醇结合蛋白4)是一种主要由肝脏分泌的小分子蛋白,属于脂质运载蛋白家族(lipocalin family),该家族成员通常具有结合和运输疏水性分子的功能。RBP4的主要生物学功能是与视黄醇(维生素A的活性形式)结合,在血液中稳定运输视黄醇至靶组织(如视网膜、上皮细胞等),从而参与视觉形成、细胞分化、免疫调节等重要生理过程。RBP4通过与甲状腺素转运蛋白(TTR)形成复合物,防止其被肾脏过滤,延长其半衰期。RBP4主要在肝脏、脂肪组织和视网膜中表达,其表达水平受维生素A状态和代谢信号的调控。RBP4基因突变可能导致其与视黄醇结合能力下降,影响维生素A的正常运输,进而引发夜盲症、干眼症等维生素A缺乏相关疾病。近年研究发现,RBP4与代谢性疾病密切相关,其血清水平在肥胖、胰岛素抵抗和2型糖尿病患者中显著升高,可能通过干扰胰岛素信号通路促进代谢紊乱。RBP4过表达会抑制脂肪细胞中葡萄糖转运蛋白GLUT4的表达,加剧胰岛素抵抗;还可能激活炎症通路,促进巨噬细胞浸润脂肪组织。相反,降低RBP4表达可改善胰岛素敏感性和糖代谢,这使其成为代谢性疾病的潜在治疗靶点。RBP4属于脂质运载蛋白家族,该家族成员具有典型的β-桶状结构,能够结合和运输脂溶性物质,如视黄醇、前列腺素、信息素等。除RBP4外,该家族还包括β-乳球蛋白、载脂蛋白D等成员,它们在免疫调节、嗅觉信号传导等过程中发挥重要作用。

This protein belongs to the lipocalin family and is the specific carrier for retinol (vitamin A alcohol) in the blood. It delivers retinol from the liver stores to the peripheral tissues. In plasma, the RBP-retinol complex interacts with transthyretin which prevents its loss by filtration through the kidney glomeruli. A deficiency of vitamin A blocks secretion of the binding protein posttranslationally and results in defective delivery and supply to the epidermal cells. [provided by RefSeq, Jul 2008]
