PTGS2(前列腺素内过氧化物合酶2),也称为COX-2,是一种编码环氧化酶-2的基因,属于前列腺素内过氧化物合酶基因家族(PTGS家族),该家族还包括PTGS1(COX-1)。PTGS家族的主要共性是催化花生四烯酸转化为前列腺素H2,这是合成多种前列腺素和血栓烷的前体分子。PTGS2主要在炎症、损伤或生长因子刺激下被诱导表达,与PTGS1(组成型表达)不同,它参与炎症反应、疼痛感知、发热及细胞增殖等过程。PTGS2的表达产物COX-2在炎症部位、肿瘤组织和某些病理状态下显著上调,是抗炎药物(如非甾体抗炎药NSAIDs)的重要靶点。PTGS2的突变可能导致其酶活性异常,与多种疾病相关,例如某些遗传性炎症疾病或癌症风险增加。PTGS2过表达会促进炎症反应、疼痛和肿瘤发生,特别是在结直肠癌等恶性肿瘤中,COX-2的高表达与肿瘤血管生成、侵袭和转移密切相关。相反,PTGS2表达降低或抑制可能减轻炎症和疼痛,但也可能影响组织修复和胃黏膜保护功能,因为某些前列腺素对胃肠道有保护作用。此外,PTGS2与其他基因如NF-κB和MAPK信号通路相关基因相互作用,调节炎症和免疫反应。PTGS2的异常表达还与心血管疾病、神经退行性疾病(如阿尔茨海默病)和关节炎等疾病有关。因此,PTGS2是炎症和癌症治疗中的重要分子靶点,但其调控需平衡治疗效果与副作用。
