NOP56(核仁蛋白56,英文全称Nucleolar Protein 56)是一种核仁蛋白,属于核糖核蛋白(RNP)家族,主要参与核糖体生物合成(ribosome biogenesis)过程。它在核仁中高度富集,与NOP58和fibrillarin等蛋白共同组成C/D box小核核糖核蛋白(snoRNP)复合物,负责指导rRNA(核糖体RNA)的2'-O-甲基化修饰,这是核糖体成熟和功能的关键步骤。NOP56通过其保守的 coiled-coil 结构域与其他蛋白相互作用,维持复合物的稳定性。若NOP56发生突变(如GGC重复扩增),可能导致其功能异常,影响rRNA加工,进而破坏核糖体组装和细胞翻译功能。这种突变与神经退行性疾病脊髓小脑共济失调36型(SCA36)密切相关,患者表现为小脑萎缩和运动协调障碍。NOP56过表达可能干扰snoRNP复合物的平衡,导致rRNA过度甲基化或核仁应激(nucleolar stress),而表达降低则可能减少核糖体生成,引发细胞增殖抑制或凋亡。NOP56所属的NOP56/NOP58基因家族成员均含有相似的蛋白互作结构域,共同参与snoRNP组装和rRNA修饰。该家族功能冗余性较低,单个基因缺陷即可显著影响核糖体合成通路。此外,NOP56的表达水平与某些癌症进展相关,可能通过调控核糖体产能影响肿瘤细胞生长。

Nop56p is a yeast nucleolar protein that is part of a complex with the nucleolar proteins Nop58p and fibrillarin. Nop56p is required for assembly of the 60S ribosomal subunit and is involved in pre-rRNA processing. The protein encoded by this gene is similar in sequence to Nop56p and is also found in the nucleolus. Multiple transcript variants encoding several different isoforms have been found for this gene, but the full-length nature of most of them has not been determined. [provided by RefSeq, Jan 2009]
