NEK8(NIMA相关激酶8)属于NEK激酶家族,该家族共有11个成员(NEK1-11),均与丝氨酸/苏氨酸激酶活性相关,参与细胞周期调控、纤毛形成和DNA损伤修复等过程。NEK8定位于纤毛基部(基体)和中心体,其激酶结构域通过磷酸化底物调控纤毛组装、维持细胞极性及有丝分裂纺锤体定位。该基因表达产物(NEK8蛋白)在肾脏发育中尤为重要,通过Hippo信号通路调控细胞增殖与分化。NEK8突变会导致纤毛结构异常,引发纤毛病(ciliopathies),如多囊肾病(ADPKD)、肾单位肾痨(nephronophthisis)和Jeune综合征(胸廓发育不良)。常见致病突变如p.Arg45Trp会破坏激酶活性,导致纤毛信号传导障碍。过表达NEK8可能激活异常细胞增殖通路(如ERK/MAPK)促进肿瘤发生,尤其在乳腺癌和肝癌中观察到其表达升高;而表达降低则导致纤毛缩短、Hedgehog信号通路失调,影响胚胎发育。NEK家族共性包括:均含保守的N端激酶结构域,通过磷酸化调控细胞周期(尤其G2/M期转换);部分成员(如NEK1/8/9)与纤毛功能相关。目前"NIMA相关激酶"的中文译名存在争议,部分文献直译为"尼马相关激酶"(原英文:NIMA-related kinase)。

This gene encodes a member of the serine/threionine protein kinase family related to NIMA (never in mitosis, gene A) of Aspergillus nidulans. The encoded protein may play a role in cell cycle progression from G2 to M phase. Mutations in the related mouse gene are associated with a disease phenotype that closely parallels the juvenile autosomal recessive form of polycystic kidney disease in humans.
