NDC80(英文全称:NDC80 kinetochore complex component)是NDC80复合体的核心组成部分,属于NDC80/HEC1基因家族。该家族成员在真核生物中高度保守,主要功能是参与染色体分离和有丝分裂过程中纺锤体微管的附着,确保遗传物质准确分配到子细胞中。NDC80复合体由NDC80、NUF2、SPC24和SPC25四个亚基组成,直接与微管结合,是连接着丝粒(kinetochore,染色体上负责牵引的结构)和纺锤体微管的关键分子桥梁。NDC80蛋白本身具有长卷曲螺旋结构域,能通过其N端与微管结合,C端则与其他复合体亚基相互作用。若NDC80发生突变(如错义突变或缺失),会导致微管附着缺陷、染色体排列错误或分离异常,引发非整倍体(染色体数目异常)和基因组不稳定性,进而促进癌症发展(如乳腺癌、结直肠癌)。NDC80在多种肿瘤中过表达,其高表达会加速细胞增殖、抑制凋亡,并与化疗耐药性相关;反之,敲低NDC80会阻滞细胞周期于有丝分裂期,诱发染色体错误分离和细胞死亡。该基因家族共性包括:均含卷曲螺旋结构、定位于着丝粒、在微管-着丝粒连接中起机械支架作用。研究还发现NDC80与BUB1、MAD2等有丝分裂检查点蛋白协同工作,确保染色体正确分离。目前针对NDC80的抑制剂(如抗微管药物)已被探索用于癌症治疗。

The encoded protein consists of an N-terminal microtubule binding domain and a C-terminal coiled-coiled domain that interacts with other components of the complex.
