MUSK(Muscle, skeletal receptor tyrosine kinase)是一种受体酪氨酸激酶,主要表达于骨骼肌神经肌肉接头处,在突触后膜的形成和维持中发挥关键作用。它的生物学功能包括参与乙酰胆碱受体(AChR)的聚集和稳定,确保神经肌肉信号的高效传递。MUSK通过与配体agrin和低密度脂蛋白受体相关蛋白4(LRP4)结合,激活下游信号通路(如Dok-7和Rapsyn),从而促进AChR的成簇化。该基因的突变会导致先天性肌无力综合征(CMS),表现为肌肉无力、易疲劳和运动障碍,因为突触后膜结构异常影响了神经肌肉传导。MUSK属于受体酪氨酸激酶(RTK)家族,该家族成员通常通过配体结合激活胞内激酶域,调控细胞生长、分化和存活。若MUSK过表达,可能增强AChR聚集,但过度激活可能干扰突触稳态;而表达降低则直接导致突触后膜发育缺陷,引发肌无力。此外,MUSK与自身免疫性疾病如重症肌无力(MG)相关,因自身抗体会靶向破坏MUSK功能。研究还发现,MUSK在癌症中可能异常表达,影响肿瘤细胞的迁移和侵袭。

This gene encodes a muscle-specific tyrosine kinase receptor. The encoded protein may play a role in clustering of the acetylcholine receptor in the postsynaptic neuromuscular junction. Mutations in this gene have been associated with congenital myasthenic syndrome. Alternatively spliced transcript variants have been described.[provided by RefSeq, Oct 2009]
