MSRB3(甲硫氨酸亚砜还原酶B3)是甲硫氨酸亚砜还原酶(MSR)基因家族的一员,属于B型亚家族,主要负责修复氧化应激导致的甲硫氨酸残基亚砜化损伤,维持蛋白质结构和功能的完整性。MSR家族成员通过催化甲硫氨酸亚砜(Met-O)还原为甲硫氨酸(Met),在抗氧化防御、细胞信号调控及衰老相关疾病中发挥关键作用。MSRB3特异性作用于R型甲硫氨酸亚砜,依赖硫氧还蛋白系统提供还原力。该基因在耳蜗毛细胞、神经系统及睾丸中高表达,尤其对听觉功能至关重要——其突变可导致常染色体隐性非综合征性耳聋(DFNB74),表现为先天性或早发性听力丧失,机制可能与毛细胞中氧化损伤积累或关键蛋白(如肌球蛋白)功能异常有关。MSRB3过表达可增强细胞抗氧化能力,减少氧化应激诱导的凋亡,但过度活跃可能干扰氧化还原信号平衡;而表达降低则使细胞对氧化损伤敏感,加速听力退化或神经退行性变。此外,MSRB3与阿尔茨海默病相关,因其能还原β-淀粉样蛋白中的氧化甲硫氨酸,抑制毒性聚集。该基因家族(MSRA/MSRB)的共性包括:依赖锌离子催化、参与衰老调控、在多种组织中介导氧化损伤修复,且与代谢性疾病、癌症及神经退行性疾病密切相关。

The protein encoded by this gene catalyzes the reduction of methionine sulfoxide to methionine. This enzyme acts as a monomer and requires zinc as a cofactor. Several transcript variants encoding two different isoforms have been found for this gene. One of the isoforms localizes to mitochondria while the other localizes to endoplasmic reticula.
