MSN(Moesin)基因属于ERM(Ezrin-Radixin-Moesin)蛋白家族,该家族成员在细胞骨架与细胞膜连接中起关键作用,参与细胞形态维持、迁移、信号传导及细胞间相互作用。MSN编码的Moesin蛋白是一种膜-细胞骨架连接蛋白,通过其N端FERM结构域与膜蛋白(如CD44、ICAMs)结合,C端与肌动蛋白(F-actin)结合,从而调控细胞极性和运动。其主要作用位点为细胞膜下的皮层区域,尤其在微绒毛、皱褶膜等结构中富集。MSN突变可能导致蛋白功能丧失或异常,引发细胞黏附缺陷、迁移障碍,与免疫缺陷(如X连锁淋巴细胞异常增生症)、癌症转移及神经发育异常相关。例如,某些突变会破坏FERM结构域与膜蛋白的结合,削弱细胞骨架重组能力。MSN过表达常见于多种肿瘤(如乳腺癌、胶质瘤),可增强肿瘤细胞侵袭性,促进EMT(上皮-间质转化);而表达降低则导致免疫细胞(如T细胞)迁移受阻,影响炎症反应。ERM家族共性包括:共享约75%氨基酸序列同源性,均含FERM-肌动蛋白结合结构域,受Rho GTP酶调控并通过磷酸化(如Thr558)激活。家族成员功能部分冗余,但组织分布不同(Moesin高表达于淋巴细胞/内皮细胞)。MSN还与HIV感染相关,因其能稳定病毒侵入所需的膜微结构。此外,MSN表达受miR-200c等调控,其异常可影响TGF-β通路,间接调控MMP9等基因表达,进一步参与肿瘤微环境重塑。
