MGA(MAX Dimerization Protein,MAX二聚化蛋白)是MYC/MAX/MAD转录调控网络中的关键成员,属于bHLH-ZIP(碱性螺旋-环-螺旋-亮氨酸拉链)蛋白家族。该家族共性是通过bHLH-ZIP结构域形成二聚体并结合DNA,调控细胞增殖、分化等过程。MGA既能与MAX蛋白形成异源二聚体结合E-box序列(DNA调控元件),又能通过T-box结构域结合T-box序列,实现双重DNA结合能力。其表达产物作为转录抑制因子,通过招募组蛋白去乙酰化酶(HDAC)等共抑制复合物,下调MYC靶基因表达,从而拮抗MYC原癌基因的促增殖作用。MGA在胚胎发育(尤其心脏和神经嵴)及维持组织稳态中起重要作用,其功能位点包括染色质调控区域和细胞周期相关基因启动子。突变可能导致功能丧失,与多种癌症相关,如结直肠癌、肺癌和淋巴瘤中常见MGA缺失或突变,导致MYC信号通路失控。若MGA过表达,会过度抑制细胞周期进程,可能引起分化异常或发育缺陷;而表达降低则削弱对MYC的拮抗作用,促进肿瘤发生。该基因与MAD家族(MXD1-4)功能相似但结构独特,因其兼具T-box和bHLH-ZIP双结构域。近期研究发现MGA突变还与CHARGE综合征(一种多发畸形疾病)相关,提示其在神经嵴细胞迁移中的非经典功能。
