乳酸脱氢酶A(LDHA)是乳酸脱氢酶家族的一员,该家族还包括LDHB、LDHC、LDHD等成员,主要功能是催化丙酮酸与乳酸之间的可逆转化,在糖酵解过程中起关键作用。LDHA在缺氧条件下尤为重要,通过将丙酮酸转化为乳酸帮助细胞在无氧环境下维持能量供应,同时再生NAD+以支持糖酵解持续进行。LDHA主要表达于骨骼肌、肝脏、心脏等代谢活跃的组织,其活性与细胞增殖、肿瘤发生密切相关。LDHA基因突变可能导致酶活性异常,例如某些错义突变会降低其催化效率,影响细胞能量代谢,与遗传性肌病、运动不耐受等疾病相关。在癌症中,LDHA常过表达,促进有氧糖酵解(瓦氏效应),为肿瘤细胞提供快速增殖所需的能量和生物合成前体,与乳腺癌、肺癌、结直肠癌等多种恶性肿瘤的进展、转移和不良预后相关。LDHA过表达还会导致微环境酸化,促进免疫逃逸,并通过调节HIF-1α等信号分子影响其他代谢相关基因的表达。相反,LDHA表达降低或抑制会迫使细胞转向氧化磷酸化,可能抑制肿瘤生长,但可能导致正常组织在缺氧条件下能量不足。LDHA抑制剂已成为抗癌研究的热点,但需注意其对正常组织尤其是肌肉活动的潜在影响。该基因家族成员均含有NAD+结合域和底物结合域,但组织分布和动力学特性略有差异,例如LDHB更倾向于催化乳酸氧化为丙酮酸,在心脏等需氧组织中占主导。

The protein encoded by this gene catalyzes the conversion of L-lactate and NAD to pyruvate and NADH in the final step of anaerobic glycolysis. The protein is found predominantly in muscle tissue and belongs to the lactate dehydrogenase family. Mutations in this gene have been linked to exertional myoglobinuria. Multiple transcript variants encoding different isoforms have been found for this gene. The human genome contains several non-transcribed pseudogenes of this gene. [provided by RefSeq, Sep 2008]
