IRAK2(白细胞介素-1受体相关激酶2)是一种丝氨酸/苏氨酸蛋白激酶,属于IRAK基因家族(包括IRAK1、IRAK2、IRAK3和IRAK4),该家族成员在先天免疫反应中起关键作用,特别是通过Toll样受体(TLR)和白细胞介素-1受体(IL-1R)信号通路传递炎症信号。IRAK家族共性在于它们均参与NF-κB和MAPK信号通路的激活,调控炎症因子(如TNF-α、IL-6)的产生。IRAK2在免疫细胞(如巨噬细胞、树突细胞)中高表达,其功能特点是作为支架蛋白和激酶双重角色:既能通过自身磷酸化激活下游信号,又能与其他适配分子(如MyD88、TRAF6)形成复合物放大信号。IRAK2的突变可能导致信号传导异常,如功能获得性突变会过度激活NF-κB,与自身免疫疾病(如类风湿关节炎)相关;而功能缺失突变可能削弱抗病原体反应,增加感染风险。研究发现IRAK2在炎症性疾病(如动脉粥样硬化)中常过表达,促进炎症因子释放,而敲低IRAK2可减轻炎症损伤。此外,IRAK2与肿瘤微环境调控相关,过表达可能通过持续激活炎症通路促进某些癌症(如乳腺癌)进展。值得注意的是,IRAK2与家族其他成员存在功能冗余,但IRAK2独特之处在于其较长的C端延长区,使其能更稳定地锚定信号复合物。在药物开发中,靶向IRAK2的小分子抑制剂正被探索用于治疗慢性炎症性疾病。
