HIPK1(Homeodomain-Interacting Protein Kinase 1)是一种丝氨酸/苏氨酸蛋白激酶,属于HIPK基因家族(包括HIPK1、HIPK2、HIPK3和HIPK4)。该家族成员共同特点是能够与同源结构域转录因子相互作用,并通过磷酸化调控多种转录因子和信号通路,参与细胞增殖、凋亡、分化及应激反应等生物学过程。HIPK1主要定位于细胞核,通过磷酸化底物蛋白(如p53、CtBP、HMGA1等)调控基因表达和细胞命运决定。在DNA损伤反应中,HIPK1可激活p53依赖的凋亡通路,但其促凋亡作用弱于家族成员HIPK2。突变或异常表达可能影响其激酶活性,导致调控功能紊乱。研究发现HIPK1在多种癌症(如乳腺癌、结直肠癌)中表达异常,其过表达可能通过激活Wnt/β-catenin等促增殖通路促进肿瘤发展,而敲低表达则可能抑制肿瘤细胞迁移侵袭能力。此外,HIPK1还参与神经发育调控,其缺失会导致小鼠运动协调障碍。该基因与糖尿病并发症也有关联,高血糖条件下HIPK1通过磷酸化PPARγ影响胰岛素敏感性。家族成员均含有保守的激酶结构域,但各亚型具有组织特异性表达模式和独特底物偏好性,形成复杂的调控网络。

The protein encoded by this gene belongs to the Ser/Thr family of protein kinases and HIPK subfamily. It phosphorylates homeodomain transcription factors and may also function as a co-repressor for homeodomain transcription factors. Alternative splicing results in four transcript variants encoding four distinct isoforms. [provided by RefSeq, Jul 2008]
