基因简述

HES5（Hairy and Enhancer of Split 5）属于HES基因家族，该家族是碱性螺旋-环-螺旋（bHLH）转录抑制因子的成员，主要参与Notch信号通路的调控。HES家族基因的共同特点是含有bHLH结构域和Orange结构域，能够通过结合DNA抑制下游靶基因的转录，在神经发育、细胞分化及维持干细胞特性中发挥关键作用。HES5主要在神经系统发育过程中表达，特别是在神经前体细胞中高表达，通过抑制神经分化相关基因（如Neurogenin和Mash1）来维持神经前体细胞的未分化状态，确保神经发生的有序进行。HES5的突变或表达异常可能导致神经发育障碍，如小头畸形或智力障碍，也可能与某些神经系统肿瘤（如神经胶质瘤）的发生相关。当HES5过表达时，会过度抑制神经分化基因，导致神经前体细胞无法正常分化为神经元或胶质细胞，可能引起发育迟缓或神经系统功能异常；而HES5表达降低则可能导致神经前体细胞过早分化，消耗干细胞池，影响神经系统的正常发育。此外，HES5还参与其他组织的发育调控，如造血系统和肠道干细胞维持。在癌症中，HES5的异常表达可能与肿瘤干细胞特性的维持有关，促进肿瘤的发生和耐药性。HES5的功能与其他HES家族成员（如HES1和HES3）有部分重叠，但各自在时空表达和靶基因选择上存在差异，共同精细调控发育过程。

The protein product of this gene, which is activated downstream of the Notch pathway, regulates cell differentiation in multiple tissues.
