GSTP1(谷胱甘肽S-转移酶P1)属于谷胱甘肽S-转移酶(GST)基因家族,这是一组参与解毒和抗氧化防御的关键酶。GST家族成员普遍具有催化谷胱甘肽(GSH)与亲电子化合物(如致癌物、药物代谢产物和氧化应激产物)结合的能力,从而促进这些有害物质的清除。GSTP1主要表达于胎盘、肺、脑等组织,尤其在肿瘤组织中常呈现异常表达。其编码的蛋白质通过结合并中和活性氧(ROS)及有毒代谢物发挥细胞保护作用,同时还能调节信号通路(如JNK通路)影响细胞增殖与凋亡。GSTP1基因的常见突变(如Ile105Val)会导致酶活性降低,削弱解毒能力,增加个体对致癌物敏感性和氧化损伤风险,这与多种癌症(如肺癌、乳腺癌、前列腺癌)的易感性相关。GSTP1过表达可能增强细胞对化疗药物的耐药性,因为许多抗癌药物正是GST酶的底物;而表达降低则会使细胞更易受氧化应激和DNA损伤的影响。此外,GSTP1的表观遗传沉默(如启动子甲基化)在前列腺癌等恶性肿瘤中频繁发生,可能促进肿瘤进展。该基因还与神经退行性疾病(如阿尔茨海默病)有关,因其抗氧化功能缺失可能加剧神经元损伤。GST家族(包括GSTA、GSTM、GSTT等亚型)的共性在于均含有保守的GSH结合域,但各亚型具有组织特异性分布和底物偏好性,共同构成机体的多层次防御系统。
