FN1基因编码纤维连接蛋白(Fibronectin),这是一种重要的细胞外基质糖蛋白,在细胞粘附、迁移、伤口修复、胚胎发育和凝血等过程中发挥关键作用。纤维连接蛋白由两个相似的亚基通过二硫键连接而成,含有多个功能域,可与胶原、肝素、纤维蛋白和整合素等分子结合,介导细胞与基质的相互作用。FN1主要在肝脏合成后分泌到血浆中,也可由成纤维细胞、内皮细胞和巨噬细胞等局部产生。FN1基因突变可能导致纤维连接蛋白功能异常,与多种疾病相关,如肾小球疾病(由于纤维连接蛋白在肾小球基底膜沉积)、纤维化疾病和某些癌症(通过促进肿瘤细胞迁移和侵袭)。FN1过表达常见于多种肿瘤,与肿瘤转移和不良预后相关,因其能增强肿瘤细胞粘附、迁移和侵袭能力;而FN1表达降低可能影响胚胎发育(导致心血管或骨骼畸形)或伤口愈合延迟。FN1属于纤维连接蛋白基因家族,该家族成员具有相似的结构特征(如III型重复序列)和功能(参与细胞-基质相互作用和组织修复)。纤维连接蛋白还通过调节TGF-β等生长因子的活性影响组织纤维化进程。在炎症反应中,FN1通过调控白细胞迁移参与免疫应答。此外,FN1的可变剪接产生多种亚型,具有组织特异性分布和功能差异。
