F2R基因也称为蛋白酶激活受体1(PAR1),是一种G蛋白偶联受体(GPCR),属于蛋白酶激活受体(PARs)家族。该家族共有4个成员(PAR1-4),其共同特点是能够被特定的蛋白酶切割激活,从而触发细胞内信号传导。F2R主要在血小板、内皮细胞、平滑肌细胞和某些神经元中表达,其激活后参与凝血、炎症、血管生成和细胞增殖等生理过程。F2R的天然激活剂是凝血酶,凝血酶切割F2R的N端,暴露出一个新的N端序列作为“栓系配体”,与受体结合并激活下游信号通路,包括Gαq、Gα12/13和Gαi介导的信号通路。F2R的突变或异常表达与多种疾病相关,例如血栓形成、动脉粥样硬化、癌症转移和炎症性疾病。F2R功能获得性突变可能导致过度凝血和血栓形成风险增加,而功能丧失性突变则可能增加出血倾向。F2R过表达与肿瘤进展和转移相关,因其促进血管生成和肿瘤细胞侵袭;而降低F2R表达可能抑制血栓形成但增加出血风险。F2R还与其他基因相互作用,例如与血管内皮生长因子(VEGF)协同促进血管生成,或与整合素相互作用影响细胞迁移。PAR家族成员的共性是它们通过蛋白水解机制激活,参与凝血、炎症和组织修复等过程,但各自有不同的组织分布和生理功能。
