ENO3(Enolase 3)是烯醇化酶家族(Enolase gene family)的重要成员之一,也被称为β-烯醇化酶或肌肉特异性烯醇化酶。该基因家族共有三个主要成员(ENO1、ENO2、ENO3),它们编码的蛋白质在糖酵解途径中催化2-磷酸甘油酸(2-PG)转化为磷酸烯醇式丙酮酸(PEP),这是产生ATP的关键步骤。ENO3主要在骨骼肌和心肌中高表达,其蛋白质产物对肌肉收缩所需的能量供应至关重要。ENO3基因突变可能导致糖酵解功能障碍,引发罕见的遗传性疾病如烯醇化酶缺乏性肌病(Enolase deficiency myopathy),患者表现为运动不耐受、肌无力甚至横纹肌溶解。研究发现ENO3表达异常与某些肌肉疾病相关,例如杜氏肌营养不良(DMD)患者肌肉组织中ENO3表达显著降低。若ENO3过表达可能增强肌肉细胞的糖酵解能力,但长期可能影响细胞代谢平衡;而表达降低则直接导致肌肉能量供应不足,影响收缩功能。除代谢作用外,ENO3还被发现具有"兼职功能"(moonlighting function),如在细胞膜表面作为纤溶酶原受体参与组织修复。该基因家族成员(ENO1/2/3)具有高度保守的活性位点,但组织分布不同:ENO1广泛表达,ENO2主要在神经元表达,而ENO3具有肌肉特异性。研究提示ENO3可能成为肌肉相关疾病的生物标志物或治疗靶点,其调控机制涉及转录因子MYOD和MEF2家族对肌肉特异性表达的调控。

This gene encodes one of the three enolase isoenzymes found in mammals. This isoenzyme is found in skeletal muscle cells in the adult where it may play a role in muscle development and regeneration. A switch from alpha enolase to beta enolase occurs in muscle tissue during development in rodents. Mutations in this gene have be associated glycogen storage disease. Alternatively spliced transcript variants encoding different isoforms have been described.[provided by RefSeq, Jul 2010]
