DLL4(Delta-like ligand 4)是Notch信号通路中的一个关键配体基因,属于Delta/Serrate/LAG-2(DSL)基因家族。该家族成员均为单次跨膜蛋白,通过结合Notch受体调控细胞间通讯,影响细胞命运决定、增殖、分化和血管生成等过程。DLL4主要在血管内皮细胞中高表达,其核心功能是调控血管发育和重塑,通过Notch信号通路介导"侧向抑制"机制,确保血管分支的适当间距和形态。当DLL4与Notch受体结合后,会触发γ-分泌酶介导的受体切割,释放Notch胞内结构域(NICD)入核调控下游靶基因。DLL4基因突变可导致多种血管发育异常,如常染色体显性遗传的Adams-Oliver综合征,表现为先天性肢体缺损和头皮缺陷。在肿瘤微环境中,DLL4过表达会促进病理性血管生成,使血管结构紊乱且通透性增加,但同时也可能通过过度激活Notch信号导致内皮细胞衰老,反而抑制肿瘤生长。降低DLL4表达或使用其抑制剂可减少肿瘤血管生成,但可能引发代偿性的血管增生。DLL4还与T细胞分化密切相关,过表达会抑制Th1细胞分化而促进调节性T细胞生成。该基因在多种癌症(如乳腺癌、胶质瘤)中异常表达,其表达水平与预后相关,已成为抗血管生成治疗的重要靶点。DSL家族成员均含有多个EGF样重复序列和DSL结构域,但各成员具有时空表达特异性和不同的受体结合偏好性,共同精细调控Notch通路的多样化功能。
