CRYZ(Crystallin Zeta)属于ζ-晶状体蛋白家族(zeta-crystallin family),该家族成员通常具有NADPH依赖的醌还原酶活性,在维持细胞氧化还原平衡中起重要作用。CRYZ主要在晶状体、肝脏和大脑中表达,其编码的蛋白质具有双重功能:一方面作为结构蛋白参与维持晶状体透明度,另一方面作为代谢酶参与解毒和抗氧化防御。CRYZ通过还原醌类化合物和α-二羰基化合物来保护细胞免受氧化损伤,这种功能对预防白内障等氧化应激相关疾病尤为重要。CRYZ基因突变可能导致酶活性丧失或结构异常,与常染色体显性先天性白内障密切相关,某些突变还会影响其代谢功能,增加氧化应激敏感性。CRYZ过表达可增强细胞抗氧化能力,但可能干扰正常代谢平衡;而表达降低则会使细胞更易受氧化损伤,加速白内障等疾病进程。该基因与醛糖还原酶(AKR1B1)功能相关,共同参与多元醇代谢通路。ζ-晶状体蛋白家族的共性在于都具有NAD(P)H结合结构域和氧化还原酶活性,在眼组织和其他器官中发挥重要的保护作用。研究表明CRYZ表达水平变化可能影响其他抗氧化基因如NQO1的表达,其功能异常还与某些神经退行性疾病和糖尿病并发症有关。

Crystallins are separated into two classes: taxon-specific, or enzyme, and ubiquitous. The latter class constitutes the major proteins of vertebrate eye lens and maintains the transparency and refractive index of the lens. The former class is also called phylogenetically-restricted crystallins. This gene encodes a taxon-specific crystallin protein which has NADPH-dependent quinone reductase activity distinct from other known quinone reductases. It lacks alcohol dehydrogenase activity although by similarity it is considered a member of the zinc-containing alcohol dehydrogenase family. Unlike other mammalian species, in humans, lens expression is low. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.
