CDIP1(细胞死亡诱导蛋白1,Cell Death-Inducing p53 Target 1)是一种由TP53基因调控的促凋亡蛋白,属于p53靶基因家族。该基因在DNA损伤或细胞应激时被激活,通过诱导细胞凋亡(程序性细胞死亡)参与肿瘤抑制。其表达产物主要定位于内质网和线粒体,通过激活内质网应激通路(如PERK/eIF2α/ATF4)和线粒体凋亡途径(如BAX/BAK依赖的细胞色素C释放)触发凋亡。CDIP1的功能特点包括与Bcl-2家族蛋白(如BAD)相互作用促进凋亡,以及通过激活caspase级联反应执行细胞死亡。突变可能导致其促凋亡功能丧失,与肿瘤发生相关,尤其在乳腺癌、结直肠癌中常见表达下调。过表达会增强细胞对凋亡信号的敏感性,可能用于癌症治疗;而低表达则可能导致细胞逃避凋亡,促进肿瘤发展。该基因属于p53靶基因家族,成员共性为受p53转录调控并参与细胞周期阻滞、DNA修复或凋亡。目前研究还发现CDIP1与神经退行性疾病(如阿尔茨海默病)中异常蛋白聚集引发的内质网应激有关,但其机制尚待深入探索。
