CCR4(趋化因子受体4,Chemokine Receptor 4)是一种G蛋白偶联受体(GPCR),属于趋化因子受体家族。该家族成员通过识别趋化因子(一类小分子信号蛋白)调控免疫细胞的迁移和激活。CCR4主要表达于Th2型辅助T细胞、调节性T细胞(Treg)和皮肤归巢T细胞表面,其配体为CCL17(TARC)和CCL22(MDC)。当配体结合后,CCR4激活下游信号通路(如MAPK和PI3K),引导免疫细胞向炎症或肿瘤部位定向迁移。功能上,CCR4参与过敏反应、自身免疫病和肿瘤微环境调控,例如在特应性皮炎中,CCR4-CCL17/22轴促进Th2细胞浸润皮肤;在成人T细胞白血病(ATL)中,CCR4常因突变(如基因融合或拷贝数变异)导致持续激活,促进癌细胞转移。若CCR4过表达,可能加剧炎症性疾病或肿瘤进展,而敲低表达会削弱Treg细胞的免疫抑制功能,可能诱发自身免疫反应。该基因属于趋化因子受体家族,其共性为具有7次跨膜结构域,通过G蛋白传递信号,调控细胞趋化性。目前针对CCR4的抗体药物(如Mogamulizumab)已用于治疗CCR4阳性淋巴瘤。专业术语中,"趋化性"指细胞沿化学梯度定向移动的能力,"Th2细胞"是分泌IL-4/IL-13的辅助T细胞亚群,"Treg"指具有免疫抑制功能的调节性T细胞。

The protein encoded by this gene belongs to the G-protein-coupled receptor family . It is a receptor for the CC chemokine - MIP-1, RANTES, TARC and MCP-1. Chemokines are a group of small polypeptide, structurally related molecules that regulate cell trafficking of various types of leukocytes. The chemokines also play fundamental roles in the development, homeostasis, and function of the immune system, and they have effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis.
