BMP7(骨形态发生蛋白7,Bone Morphogenetic Protein 7)属于TGF-β超家族中的BMP亚家族,是一类分泌型信号分子,主要调控胚胎发育、骨骼形成、组织修复及细胞分化等过程。其表达产物为一种二聚体蛋白,通过激活SMAD信号通路(一类细胞内传递信号的蛋白质)发挥作用,主要靶向间充质干细胞(未分化的多能细胞),诱导其分化为成骨细胞(骨形成细胞)或软骨细胞。BMP7在肾脏、眼睛和骨骼中表达较高,尤其在肾脏发育中至关重要,可抑制纤维化(组织异常瘢痕化)并促进上皮细胞再生。若BMP7发生功能丧失性突变(如错义突变或缺失),可能导致骨骼畸形(如短指症)、先天性肾脏缺陷或眼部异常;而某些获得性功能突变可能与肿瘤发生相关,如促进某些癌症的转移。BMP家族(含BMP2、BMP4等)的共性是通过结合细胞膜上的I/II型受体激活下游信号,调控细胞增殖、分化和凋亡(程序性细胞死亡)。当BMP7过表达时,可能加速骨修复但也会导致异位骨化(非骨骼部位异常骨形成),或抑制肿瘤生长;而低表达则与慢性肾病进展、骨质疏松或角膜损伤修复延迟相关。在疾病方面,BMP7下调与糖尿病肾病、肺纤维化相关,而上调在某些骨肉瘤中可能促进侵袭性。目前重组BMP7蛋白已用于临床治疗骨折和脊柱融合,但需注意剂量以避免副作用。

The bone morphogenetic proteins (BMPs) are a family of secreted signaling molecules that can induce ectopic bone growth. Many BMPs are part of the transforming growth factor-beta (TGFB) superfamily. BMPs were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. Based on its expression early in embryogenesis, the BMP encoded by this gene has a proposed role in early development and possible bone inductive activity. [provided by RefSeq, Jul 2008]
