AGTR2(血管紧张素II受体2型)属于血管紧张素受体家族,主要参与调节心血管和肾脏功能。它通过结合血管紧张素II(Ang II)发挥作用,但其作用与同家族的AGTR1(1型受体)相反,通常具有抗增殖、抗纤维化和血管舒张作用。AGTR2主要分布在胎儿组织、肾上腺髓质、子宫、卵巢、心脏和肾脏中,在成年后表达水平较低。该基因的突变可能导致受体功能异常,影响血压调节、血管重塑和细胞增殖,与高血压、动脉粥样硬化、心力衰竭和糖尿病肾病等疾病相关。AGTR2过表达可能通过抑制AGTR1的作用减轻心血管疾病进展,而降低表达则可能加剧血管收缩和纤维化。AGTR2还参与神经保护、炎症调节和胚胎发育,其激活可促进一氧化氮(NO)释放,改善内皮功能。在基因家族方面,AGTR2属于G蛋白偶联受体(GPCR)超家族中的血管紧张素受体亚家族,与AGTR1共享配体(Ang II)但信号通路不同(AGTR2主要通过Gi/o和Gq通路)。该家族成员均含7个跨膜结构域,通过调节血压、体液平衡和细胞生长维持内环境稳定。研究表明AGTR2基因多态性与心血管疾病风险相关,其表观遗传调控也可能影响疾病易感性。
