ACTN3(α-actinin-3)是一种编码α-辅肌动蛋白3的基因,属于肌动蛋白结合蛋白家族(actin-binding protein family),主要表达于骨骼肌的快肌纤维(fast-twitch fibers)中。其蛋白产物α-actinin-3通过与肌动蛋白(actin)结合,参与维持肌肉收缩的结构基础,尤其在需要爆发力的运动中(如短跑、举重)发挥关键作用。该基因的生物学功能包括稳定肌纤维的Z盘结构(Z-disc,肌肉收缩的基本单位)和调节肌肉代谢效率。ACTN3基因最常见的突变是R577X(第577位的精氨酸变为终止密码子),导致蛋白翻译提前终止,产生无功能的截短蛋白。约18%的亚洲人和非洲人携带这种纯合突变(XX型),表现为α-actinin-3完全缺失,但通常不会引起疾病,仅与运动表现相关:缺失者爆发力较弱,但耐力运动可能更具优势。研究发现,ACTN3 XX基因型与低肌肉强度、骨质疏松风险增加相关,也可能影响神经肌肉疾病(如杜氏肌营养不良)的严重程度。若该基因过表达,可能增强快肌纤维功能,提升无氧运动能力;而表达降低时,肌肉更容易转向慢肌纤维(slow-twitch fibers)特性,更适合耐力活动。ACTN3属于α-actinin基因家族,该家族共有4个成员(ACTN1-4),共性在于含有保守的肌动蛋白结合域和钙调蛋白样结构域,功能均与细胞骨架组织、细胞黏附及信号传导相关。其中ACTN1/4广泛分布于非肌肉细胞,而ACTN2/3特异性存在于肌肉组织。目前"α-actinin-3"的中文译名存在"α-辅肌动蛋白3"和"α-肌动蛋白结合蛋白3"等不同表述,建议统一使用前者。

This gene encodes a member of the alpha-actin binding protein gene family. The encoded protein is primarily expressed in skeletal muscle and functions as a structural component of sarcomeric Z line. This protein is involved in crosslinking actin containing thin filaments. An allelic polymorphism in this gene results in both coding and non-coding variants; the reference genome represents the coding allele.
