The CHIA gene, also designated as Chitinase Acidic or Chitinase 1, encodes a member of the chitinase family, a group of enzymes characterized by their ability to hydrolyze chitin, a structural polysaccharide prevalent in the cell walls of fungi, the exoskeletons of insects, and the integuments of certain parasites. Expressed predominantly in macrophages and epithelial cells, CHIA plays a pivotal role in innate immune defense by localizing to the extracellular matrix and infection sites, where it degrades chitinous components of pathogens to compromise their structural integrity and facilitate their clearance. The enzymatic activity of CHIA is tightly regulated by various immunological stimuli, including cytokines such as interferon-gamma and pathogen-associated molecular patterns (PAMPs), allowing it to adapt its function during inflammatory responses and tissue remodeling. While this activity is essential for protecting the host against fungal and parasitic infections, dysregulation of CHIA expression is implicated in several chronic inflammatory conditions, including asthma, chronic obstructive pulmonary disease (COPD), and inflammatory bowel disease (IBD). Specifically, mutations that reduce or abolish enzyme activity can impair the body's defensive capacity against pathogens, increasing susceptibility to infection, whereas overexpression, although potentially enhancing pathogen clearance, may exacerbate inflammatory cascades and lead to collateral tissue damage. Consequently, CHIA represents a potential therapeutic target for modulating immune responses in both infectious and inflammatory diseases, although its precise clinical application remains an area of ongoing investigation.
