NAPA基因编码N-乙基马来酰亚胺敏感因子附着蛋白α(α-SNAP),属于SNAP蛋白家族。该家族成员在细胞内囊泡运输和膜融合过程中起关键作用,特别是参与SNARE复合体的组装与解离。α-SNAP作为通用适配器蛋白,通过与NSF(N-乙基马来酰亚胺敏感因子)及SNARE蛋白相互作用,促进SNARE复合体的解离,从而保证囊泡运输的可持续性。其主要作用位点包括突触小泡、高尔基体及内体等膜性细胞器。该基因突变会导致功能异常,与神经退行性疾病如婴儿神经轴索营养不良(INAD)密切相关,表现为运动障碍、智力退化等症状,因突变更扰乱了神经元内囊泡运输。若NAPA过表达,可能增强囊泡运输效率但可能导致膜融合失控;而表达降低则会损害突触传递等依赖囊泡运输的过程,影响神经递质释放。SNAP蛋白家族(包括α/β/γ-SNAP)的共性在于均含有保守的SNAP核心结构域,能结合NSF和SNARE蛋白,但组织分布与亲和力存在差异。研究还发现α-SNAP与自噬溶酶体通路及某些病毒感染(如乙脑病毒)的膜融合过程相关,其表达水平变化可能通过这些途径影响疾病进展。

This gene encodes a member of the soluble NSF attachment protein (SNAP) family. SNAP proteins play a critical role in the docking and fusion of vesicles to target membranes as part of the 20S NSF-SNAP-SNARE complex. The encoded protein plays a role in the completion of membrane fusion by mediating the interaction of N-ethylmaleimide-sensitive factor (NSF) with the vesicle-associated and membrane-associated SNAP receptor (SNARE) complex, and stimulating the ATPase activity of NSF.
