GIPR(葡萄糖依赖性促胰岛素多肽受体)属于B类G蛋白偶联受体(GPCR)家族,该家族成员通常通过结合肽类激素激活细胞内信号通路。GIPR主要表达于胰腺β细胞、脂肪细胞和某些脑区,其天然配体是葡萄糖依赖性促胰岛素多肽(GIP)。GIPR的主要功能是介导GIP的生物学效应,包括促进葡萄糖刺激的胰岛素分泌(肠促胰岛素效应)、抑制胃酸分泌、促进脂肪生成和骨形成。在胰腺β细胞中,GIPR激活后通过Gs蛋白-cAMP-PKA通路增强胰岛素分泌,这一作用高度依赖血糖水平。GIPR基因突变可能导致功能丧失或获得,功能丧失型突变与2型糖尿病风险增加相关,而持续激活突变可能引起低血糖。GIPR过表达可能增强胰岛素分泌和脂肪储存,与肥胖相关;而表达降低则可能削弱肠促胰岛素效应,导致血糖控制受损。在2型糖尿病患者中常观察到GIPR信号通路敏感性下降("GIP抵抗")现象。GIPR与GLP-1R同属肠促胰岛素受体家族,两者协同调节血糖,但GIPR更偏向促进能量储存。目前GIPR/GLP-1R双重激动剂(如替尔泊肽)已成为新型糖尿病/肥胖治疗药物,通过同时激活两个受体产生协同降糖减重效果。该基因的遗传变异与代谢综合征、骨质疏松等疾病易感性相关。

This gene encodes a G-protein coupled receptor for gastric inhibitory polypeptide (GIP), which was originally identified as an activity in gut extracts that inhibited gastric acid secretion and gastrin release, but subsequently was demonstrated to stimulate insulin release in the presence of elevated glucose. Mice lacking this gene exhibit higher blood glucose levels with impaired initial insulin response after oral glucose load. Defect in this gene thus may contribute to the pathogenesis of diabetes.
