FGF20(成纤维细胞生长因子20)属于FGF基因家族,该家族包含22个成员,主要参与细胞增殖、分化、迁移和存活等生物学过程。FGF20在胚胎发育、组织修复和代谢调节中发挥重要作用,尤其在神经系统和毛囊发育中表现突出。FGF20通过激活FGFR受体(通常是FGFR1c和FGFR3c)和下游MAPK、PI3K/AKT等信号通路发挥作用,主要表达于大脑(如黑质和纹状体)、肾脏和皮肤等组织。FGF20的突变可能导致功能丧失,与帕金森病风险增加相关(如rs1721100多态性),还可能影响毛囊发育导致脱发。该基因过表达可能促进肿瘤发生(如胶质瘤和乳腺癌),因其刺激细胞增殖和血管生成;而表达降低则可能损害神经保护功能或导致发育异常。FGF家族成员的共性是依赖肝素/硫酸乙酰肝素结合来激活受体,多数以旁分泌方式发挥作用(FGF15/19/21/23除外)。FGF20与家族成员FGF9和FGF16具有高度同源性,三者构成FGF9亚家族,均参与器官形成。研究还发现FGF20能与其他生长因子(如WNT)协同调控毛囊周期,其表达异常可能破坏这种平衡。
