C1S基因(补体成分1亚成分s,英文:Complement C1s)属于补体系统(Complement system)的C1复合物基因家族,该家族还包括C1R和C1Q基因。补体系统是先天免疫的重要组成部分,通过“补体级联反应”识别并清除病原体或异常细胞。C1S编码的蛋白是一种丝氨酸蛋白酶(serine protease,能切割其他蛋白质的酶),在经典补体激活途径中起关键作用。C1S与C1R共同形成C1复合物的催化核心,当C1复合物结合抗体-抗原复合物(如细菌表面的抗体标记)时,C1S被激活,进而切割下游补体成分C4和C2,启动补体级联反应,最终形成“膜攻击复合物”(MAC)直接裂解病原体,或通过调理作用(opsonization,标记病原体以便免疫细胞吞噬)增强清除效率。C1S的突变可能导致补体功能缺陷,例如遗传性血管性水肿(HAE)或系统性红斑狼疮(SLE)等自身免疫病,因补体过度激活或调控失衡引发炎症和组织损伤。C1S过表达可能加剧补体介导的炎症反应,与类风湿关节炎或缺血再灌注损伤相关;而表达降低则增加感染风险或导致免疫复合物清除障碍。C1S基因家族成员均参与补体激活,具有类似蛋白酶功能的结构域(如丝氨酸蛋白酶结构域),但各自作用于不同靶点。C1S的活性受C1抑制剂(C1-INH)严格调控,突变若影响其与抑制剂的结合,可能导致不受控的补体激活。研究还发现C1S在肿瘤微环境中可能促进癌症进展,因其参与细胞外基质重塑(extracellular matrix remodeling,影响组织结构的动态过程)。

This gene encodes a serine protease, which is a major constituent of the human complement subcomponent C1. C1s associates with two other complement components C1r and C1q in order to yield the first component of the serum complement system. Defects in this gene are the cause of selective C1s deficiency.
