IFNB1基因编码干扰素β(Interferon beta,IFN-β),是一种重要的细胞因子,属于I型干扰素家族(包括IFN-α、IFN-β、IFN-ω等)。该家族成员具有抗病毒、抗增殖和免疫调节功能,通过激活JAK-STAT信号通路诱导干扰素刺激基因(ISGs)的表达。IFN-β主要由成纤维细胞和免疫细胞产生,通过结合干扰素α/β受体(IFNAR)发挥作用,主要靶向免疫细胞(如树突细胞、巨噬细胞)和病毒感染细胞。其生物学功能包括抑制病毒复制、增强MHC I类分子表达以促进抗原呈递、调节Th1/Th2免疫平衡以及抑制某些肿瘤细胞增殖。IFNB1基因突变可能导致干扰素信号通路异常,与多种疾病相关,如突变导致功能丧失可能增加病毒感染易感性,而某些单核苷酸多态性(SNPs)与多发性硬化症(MS)易感性相关。IFN-β是多发性硬化症的一线治疗药物,因其能抑制中枢神经系统炎症反应。IFNB1过表达会增强抗病毒和抗肿瘤免疫,但可能导致自身免疫反应(如流感样症状);而表达降低则可能削弱宿主防御能力,增加感染或肿瘤风险。该基因家族(I型干扰素)的共性包括:依赖IFNAR受体、激活相似的信号通路、具有重叠但非冗余的生物学功能,以及共享抗病毒和免疫调节特性。
