ACSL1(长链脂酰辅酶A合成酶1)属于ACSL基因家族,该家族共有5个成员(ACSL1、ACSL3、ACSL4、ACSL5、ACSL6),主要功能是催化长链脂肪酸(如棕榈酸、油酸等)与辅酶A结合形成脂酰辅酶A,这是脂肪酸代谢的关键步骤,参与甘油三酯合成、β-氧化和膜磷脂合成等过程。ACSL1在肝脏、脂肪组织、心脏和骨骼肌中高表达,尤其在脂肪组织中起重要作用,负责将游离脂肪酸转化为可储存或利用的形式。ACSL1的突变可能导致其酶活性降低,影响脂肪酸代谢,与肥胖、胰岛素抵抗和2型糖尿病相关。研究表明,ACSL1过表达会增加细胞内脂质积累,促进脂肪生成,可能加剧代谢紊乱;而降低ACSL1表达会减少脂肪储存,改善胰岛素敏感性,但也可能影响能量供应。ACSL1还与炎症反应有关,其过表达可能通过增加促炎因子(如TNF-α、IL-6)的释放加剧代谢性疾病。此外,ACSL1在癌症中也有研究,某些肿瘤中其表达升高可能与癌细胞依赖脂质代谢有关。ACSL基因家族的共性在于它们都含有高度保守的ATP/AMP结合域和脂肪酸结合域,但各成员在组织分布和底物偏好上略有差异,例如ACSL1偏好碳链长度为16-18的脂肪酸。总体而言,ACSL1是脂质代谢的核心调控基因,其表达失衡可能引发多种代谢疾病,是潜在的治疗靶点。
