ACOX2(酰基辅酶A氧化酶2,英文全称Acyl-CoA Oxidase 2)是一种参与过氧化物酶体β-氧化途径的关键酶,主要负责催化超长链脂肪酸(VLCFAs)和支链脂肪酸(如胆汁酸中间体)的氧化分解。其表达产物通过生成过氧化氢(H2O2)和缩短的酰基辅酶A,为细胞提供能量并参与脂质代谢平衡。ACOX2主要作用位点是过氧化物酶体(一种细胞器),其功能缺陷会导致有毒代谢物积累,引发神经退行性疾病或肝功能障碍。突变可能造成酶活性丧失,导致过氧化物酶体紊乱(如齐薇格谱系障碍,Zellweger spectrum disorder),表现为发育迟缓、肝肿大或神经系统异常。ACOX2与某些遗传代谢病(如胆汁酸合成障碍)相关,其表达异常可能干扰脂溶性维生素吸收或引发炎症反应。若ACOX2过表达,可能增加氧化应激(细胞损伤主因之一)风险;而表达降低则导致脂肪酸代谢受阻,诱发脂肪肝或神经损伤。ACOX2属于酰基辅酶A氧化酶家族(ACOX家族),该家族共性为均含FAD(黄素腺嘌呤二核苷酸)辅基,通过氧化反应参与脂肪酸降解。家族成员(如ACOX1、ACOX3)分工不同,但均与脂质稳态密切相关。

The product of this gene belongs to the acyl-CoA oxidase family. It encodes the branched-chain acyl-CoA oxidase which is involved in the degradation of long branched fatty acids and bile acid intermediates in peroxisomes. Deficiency of this enzyme results in the accumulation of branched fatty acids and bile acid intermediates, and may lead to Zellweger syndrome, severe mental retardation, and death in children.
