ABCA7属于ATP结合盒转运蛋白(ABC转运蛋白)超家族中的A亚家族,该家族成员以依赖ATP的能量跨膜转运脂质和其他疏水分子为主要功能。ABCA7在人体中主要表达于大脑、免疫系统和巨噬细胞中,其编码的蛋白质通过促进磷脂和胆固醇的转运参与细胞膜稳态维持及脂质代谢调控。该基因在阿尔茨海默病(AD)中具有重要关联,其功能缺失突变或表达降低可能导致β-淀粉样蛋白(Aβ)清除能力下降,加速淀粉样斑块沉积。全基因组关联研究(GWAS)发现ABCA7的某些变异(如rs3764650)显著增加晚发性AD风险,尤其在非裔人群中风险效应更突出。此外,ABCA7通过调节吞噬功能影响免疫反应,其表达水平变化可能改变巨噬细胞对凋亡细胞的清除效率。在ABCA7基因敲除小鼠模型中观察到Aβ积累增加和认知功能缺陷,而过表达实验显示其可能通过增强载脂蛋白E(apoE)的脂化作用促进Aβ降解。该基因还与高密度脂蛋白(HDL)代谢相关,但具体机制不同于家族明星成员ABCA1。ABCA7的异常表达还涉及其他神经系统疾病和自身免疫性疾病,如多发性硬化症。基因家族共性方面,ABC转运蛋白均含有特征性的核苷酸结合结构域(NBD)和跨膜结构域(TMD),利用ATP水解产生的能量实现底物跨膜转运,参与药物抵抗、抗原呈递等多种生理过程。需要注意的是,ABCA7的功能具有物种差异性,小鼠模型中某些表型与人类疾病关联仍需进一步验证。目前针对ABCA7的治疗策略主要集中在调节其表达水平以改善AD病理,但尚未进入临床阶段。
